Research progress on the application of antimicrobial peptides in the detection of foodborne pathogenic bacteria
Foodborne diseases caused by pathogenic bacteria are a serious threat to human health and a serious public health problem. Rapid and accurate detection of foodborne pathogenic bacteria in food is important for the prevention and control of foodborne diseases. In recent years, a variety of different highly sensitive and specific rapid detection methods have been developed with the aim of realizing the rapid detection of pathogenic bacteria in food. These methods all need to specifically identify bacteria from complex food matrixes. To achieve this goal, many small molecule substances have been developed, such as antibodies, aptamers, phages, antimicrobial peptides (AMPs), lectins, and enzymes. These small molecule substances have different advantages and disadvantages in application. The application of antimicrobial peptides has received more and more attention because of their high stability, simple synthesis, easy availability, high affinity for bacteria, low cost of AMPs compared with antibodies, DNA probes, more stable in harsh environments, and easy to be mass-produced. Therefore, they are being applied to the development and utilization of a variety of rapid detection methods. In this review, we systematically summarized the application of antimicrobial peptides in the detection of foodborne pathogens, and discussed its application prospects.

Effect and mechanism of dapagliflozin on pulmonary arterial hypertension induced by monocrotaline in rats
Objective To investigate the effect of dapagliflozin (DAPA) on monocrotaline (MCT)-induced pulmonary arterial hypertension (PAH) in rats and its mechanism of action. Methods Thirty male SD rats were randomly divided into three groups: (1) Control group, (2) Model group: pulmonary arterial hypertension group, (3) DAPA group: dapagliflozin treatment group. Single subcutaneous injection of MCT (60 mg/kg) was administered to establish the PAH model. The control group and the MCT group were given 0.5% sodium carboxymethyl cellulose by gavage every day, while the DAPA group was given dapagliflozin (1 mg/kg) dissolved in 0.5% sodium carboxymethyl cellulose by gavage. After 21 days, the right ventricular systolic pressure (RVSP) and right ventricular hypertrophy index (RVHI) of rats were measured, the percentage of pulmonary arteriolar wall thickness was measured by HE staining, pulmonary arterial α-smooth muscle actin was detected by immunohistochemistry, NLRP3, IL-1β, and IL-18 protein expression in lung tissue were detected by Western blot, and plasma IL-1β and IL-18 levels were measured by Elisa. Results DAPA significantly reduced RVSP and improved right ventricular hypertrophy in MCT-induced PAH rats, inhibited the thickening of pulmonary arterioles and decreased the degree of muscularization in PAH rats. Furthermore, DAPA inhibited the activation of the NLRP3 inflammasome in lung tissue and down-regulated the levels of IL-1β and IL-18 in lung tissue and plasma. Conclusion DAPA can reduce the RVSP, improve right ventricular hypertrophy and pulmonary vascular remodeling in MCT-induced PAH rats, which may be associated with its inhibition of NLRP3 inflammasome activation and downregulation of IL-18 and IL-1β expression levels.

Preparation of succinylated antibody to the K89 position of AIF and study of the interacting proteins with AIF
Objective Our research team has previously found that AIF protein exhibits increased levels of lysine 89 succinylation modification in non-smoking female lung adenocarcinoma patients. Our team aims to prepare an antibody that can recognize the succinylation modification of lysine 89 residue on AIF, which will be used to detect AIF succinylation at this site. Methods Bioinformatics analysis was conducted using online databases to analyze the physicochemical properties, transmembrane structure, signal peptide, protein structure, and succinylation sites of AIF protein. Based on the amino acid sequence of AIF, three peptides were synthesized, including the lysine 89 residue (K89) site. Among them, two peptides containing AIF K89 were succinylated. The three synthesized peptides were coupled with KLH and then immunized in rabbits to prepare antibodies specifically recognizing AIF K89 succinylation modification. The antibody efficacy and specificity were evaluated using ELISA, Dot blot, Western blotting, and immunohistochemistry. Gel digestion and high-resolution mass spectrometry were used to analyze the proteins interacting with AIF. Results Bioinformatics analysis revealed that succinylation modification of AIF K89 led to a change in the α-helical structure formed by amino acid residues 79-99 of AIF: it transformed from a longer α-helix into two shorter α-helices with a gap. The two synthesized succinylated peptides and one non-succinylated peptide successfully generated corresponding antibodies in rabbits. The antibody titers in the immune sera detected by ELISA were 1: 486, 000, 1: 162, 000, and 1: 18, 000, respectively. The prepared antibody recognizing AIF K89 succinylation modification exhibited 27 times higher efficacy in recognizing succinylated peptides compared to non-succinylated peptides, as determined by antibody Dot blot. Western blotting results showed that the AIF succinylation antibody specifically recognized the AIF K89 succinylation modification site. Mass spectrometry identified proteins interacting with AIF, including XIAP/BIRC4, EIF3G, and PRELID1. Conclusion Bioinformatics analysis suggests that succinylation modification of AIF K89 affects the structure of AIF protein. The prepared AIF K89 succinylation antibody can specifically recognize AIF K89 succinylation modification, providing a foundation for further investigation into the mechanism of AIF succinylation.

Down-regulating SCD1 expression sensitizes RSL3 induced ferroptosis and inhibited bladder cancer cell growth
Objective To explore the effect and mechanism of SCD1 protein expression on the ability of ferroptosis activator RSL3 to induce ferroptosis in bladder cancer cells. Method T24 cells were treated with SCD1 small interfering RNA (siRNA), specific SCD1 inhibitor (A9) or combined with RSL3; The effects of inhibiting SCD1 expression or combining with RSL3 on cell proliferation and clonogenesis were detected by CCK-8 and clonogenesis assay; Malondialdehyde (MDA) and lipid peroxides (LPO) were determined to investigate the effect of down-regulated SCD1 on the ability of RSL3 to induce ferroptosis in bladder cancer cells T24; Finally, glutathione peroxidase 4(GPX4) expression was detected by western blot. Results Both siRNA silencing and A9 treatment decreased the proliferation and colony formation ability of bladder cancer cells. Compared with the single treatment group, The two SCD1 inhibition methods combined with RSL3 further increased the MDA and LPO content in the cells, and enhanced the degree of inhibition on the proliferation and clonal colony formation of bladder cancer cells. In addition, inhibition of SCD1 enhanced the inhibitory effect of RSL3 on GPX4 protein expression. Conclusion Down-regulation of SCD1 can inhibit the growth of bladder cancer cells by enhancing the inhibition of GPX4 protein expression by RSL3 and inducing ferroptosis.

Objective This study aimed to analyze the factors influencing improvement in prostate symptoms in patients with benign prostatic hyperplasia (BPH) treated with either bipolar transurethral resection of the prostate (BPRP) or holmium laser enucleation of the prostate (HoLEP). Methods We conducted a retrospective analysis of data from 240 patients diagnosed with BPH and treated between October 2018 and October 2021 at Tongji Hospital, Wuhan. Preoperative and postoperative data were reviewed, including urodynamic parameters in some patients, International Prostate Symptom Score (IPSS), Overactive Bladder (OAB) symptoms, Quality of Life (QoL) scores, surgical approach, and perioperative information. A telephone follow-up was performed at 6 months post-surgery to record relevant follow-up data. Among the 240 patients, 124 completed urodynamic examinations, and data from these patients were statistically analyzed to explore factors influencing postoperative improvement in prostate symptoms. Improvement in IPSS was defined as (postoperative/preoperative) ≤0.50, and patients were divided into the Improvement Group (Group A, 190 cases) and the Poor Improvement Group (Group B, 50 cases). Results All 240 patients who met the inclusion criteria had successful surgeries, with 178 patients undergoing HoLEP and 62 patients undergoing BPRP. Age, disease duration, BMI, FBG, TC, TG, AST, ALT, ALB, Csr, GFR, postoperative IPSS, preoperative and postoperative QoL, preoperative and postoperative OAB, surgical duration, indwelling catheter time, length of hospital stay, preoperative urinary retention, history of cardiovascular disease, history of cerebral infarction, history of diabetes, surgical approach, Qmax, FDV, PVR, Pqmax, SDV, BOOI, and BCI were not identified as risk factors for poor postoperative IPSS improvement. Prolonged disease duration (P<0.001), prostate volume (PV) (P=0.024), total prostate-specific antigen (TPSA) (P=0.002), free prostate-specific antigen (FPSA) (P=0.006), elevated preoperative IPSS (P=0.015), and decreased hemoglobin (Hb) (P=0.016) were identified as risk factors for poor postoperative IPSS improvement, with preoperative IPSS (P=0.006) and elevated PV (P=0.035) being independent risk factors. Conclusion Patients with larger prostate volumes and higher preoperative IPSS scores may experience poorer IPSS improvement after BPRP or HoLEP treatment for BPH. These patients should be counseled before surgery regarding the possibility of persistent prostate symptoms postoperatively, and medication may be necessary to alleviate symptoms if required.

The clinical data, PI3K/AKT signaling pathway markers [PI3K, phosphorylated AKT (p-AKT) ], fasting blood glucose (FPG), insulin resistance index (Homa - insulin resistance, HOMA-IR) were compared between the two groups, and the effects of PI3K and p-AKT on abnormal glucose tolerance in women during pregnancy were analyzed by interaction. Pearson correlation coefficient was used to analyze the correlation between PI3K and p-AKT levels and FPG and HOMA-IR in pregnant women with abnormal glucose tolerance during pregnancy. The clinical data and serum PI3K, p-AKT, FPG levels and HOMA-IR of pregnant women with simple glucose tolerance abnormalities and pregnant women with gestational diabetes mellitus (GDM) in the study group were compared, and the factors influencing GDM were analyzed by logistic multi-factor regression model, and the value of serum PI3K and p-AKT levels in predicting GDM was analyzed by subject work characteristic (ROC) curve. RESULTS: Serum PI3K levels were lower in the study group than in the control group, and serum p-AKT, FPG levels and HOMA-IR were higher than in the control group; low PI3K levels and high p-AKT levels showed a positive interaction in a superphase multiplicative model of glucose tolerance abnormalities in pregnant women; serum PI3K levels in pregnant women with glucose tolerance abnormalities in pregnancy were negatively correlated with FPG, HOMA-IR, and p-AKT levels were negatively correlated with FPG and HOMA-IR. Body mass index, family history of diabetes, serum p-AKT, FPG levels and HOMA-IR were higher in pregnant women with GDM than in pregnant women with simple glucose tolerance abnormalities, and serum PI3K levels were lower than in pregnant women with simple glucose tolerance abnormalities; body mass index, family history of diabetes, serum PI3K and p-AKT levels were all influencing factors for GDM; serum The area under the curve (AUC) for the prediction of GDM by serum PI3K and p-AKT levels were all>0.7, and the AUC for the combined prediction of the two was the largest at 0.916, and the sensitivity and specificity of the prediction were 90.62% and 80.13%, respectively. Conclusion Changes in the levels of PI3K and p-AKT, markers of PI3K/AKT signaling pathway, are closely associated with abnormal glucose tolerance and insulin resistance in women during pregnancy, and their combination has good predictive efficacy in predicting GDM.

The expression and clinical significance of HOXC8 in colon cancer
Objective To investigate the expression and clinical significance of Homeobox C8 (HOXC8) in colon cancer (CC) . Methods The Cancer Genome Atlas (TCGA) database and GEO database were used to analyze HOXC8 mRNA expression in CC tissues. A total of 100 patients with CC tissue specimens and clinicopathological data were collected from Chongqing Medical University First Affiliated Hospital, and HOXC8 protein was detected by immunohistochemistry. Moreover, the association between HOXC8 expression and clinicopathological characteristics was analyzed. Univariate and multivariate Cox regression models and Kaplan-Meier were used to evaluate the prognostic value of HOXC8 in CC. The GEPIA database was used to evaluate the correlation between HOXC8 and immune cell biomarkers. Results Multi databases analysis and detection of patient tissue samples showed that the mRNA and protein expression level of HOXC8 is increased in cancer tissue (P<0.05, P<0.001) . In addition, the high expression of HOXC8 is related to poor overall survival rate (In TCGA data set, P<0.001; In GEO GSE39582 data set, P=0.008) . Cox regression analysis suggests that HOXC8 is an independent risk factor affecting the prognosis of patients with CC (In TCGA data set, P=0.002; In GEO GSE39582 data set, P= 0.039) . We also studied the potential role of HOXC8 in tumor-immune interactions, and found that HOXC8 may be significantly associated with multiple immune cells in colon cancer (P< 0.001) . Conclusion Our study showed that the expression of HOXC8 is up-regulated in CC, and revealed the prognostic potential of HOXC8 in CC and its potential as an immune-related therapeutic target for CC.

Clinical efficacy of atorvastatin calcium tablets combined with sacubitril/valsartan in patients with chronic heart failure
Objective The aim of this report is to explore the effects of atorvastatin calcium tablets combined with sacubitril/valsartan on clinical efficacy, cardiac color ultrasound indexes and serum indexes in patients with chronic heart failure. Methods We collected data of 87 patients with chronic heart failure admitted to Sanzhen Rehabilitation Hospital in Changsha, Hunan Province from January 2022 to December 2022. According to the random number table method, the patients were divided into control group (treated with atorvastatin calcium tablets orally and diuretics, etc. ) with 43 cases and observation group (treated with sacubitril/valsartan plus the same therapy as the control group) with 44 cases. Results were compared between the two groups, including post-therapy efficacy and readmission rates, cardiac ultrasound and serum indexes before and after treatment, and the occurrence of adverse reactions during treatment. Results After 2 months of treatment, the total clinical efficiency of the observation group was 95.45%, which was significantly higher than that of the control group (74.42%). And the overall incidence of adverse reactions and the readmission rate of the observation group were observably lower than those of the control group. Compared with the pre-therapy period, an increase of various indicators was significantly obvious in both groups after treatment, including left ventricular ejection fraction (LVEF), systolic peak velocity (SPV), left anterior descending branch diastolic peak velocity (DPV) and time velocity integral (TVI). And the changes in the observation group were signally more obvious than the control group, with statistically significant differences between the two groups. Both of the two groups had an evident decrease in the left ventricular end-systolic diameter (LVESD) and the left ventricular end-diastolic diameter (LVEDD). And the LVESD and LVEDD were lower in the observation group than in the control group. However, there was no statistically significant difference in LVEDD level through the intra-group comparison of the control group before and after treatment. Conclusion Atorvastatin calcium tablets combined with sacubitril/valsartan has a good application effect in patients with chronic heart failure, as it apparently contributes to improving the prognosis and enhancing the quality of their lives.

Effect of single-port thoracoscopic segmentectomy on lung function, and disease free survival in patients with early NSCLC
Objective To explore the effect of single-port thoracoscopic segmentectomy on lung function, serum trauma response and long-term metastasis in patients with early non-small cell lung cancer (NSCLC). Methods 200 patients with early NSCLC admitted to our hospital from April 2020 to September 2021 were divided into the control group (100 cases) and observation group (100 cases) by using a computer to randomly generate numbers. The control group received single-port thoracoscopic lobectomy while the observation group patients recceived single-port thoracoscopic segmentectomy. The surgical conditions of the two groups was compared, including lung function [forced vital capacity (FVC), forced expiratory volume in one second (FEV1) and FEV1/FVC ratio], serum trauma response indicators [prostaglandin (PGE2), cortisol (Cor) and C-reactive protein (CRP) ] and tumor cell carcinoembryonic antigen (CEA), cytokeratin 19 fragment (CYFRA21-1) and squamous cell carcinoma antigen (SCC-Ag). The survival and long-term metastasis were tested by Kaplan-Meier curve, and the complications were compared. Results Compared with control group, the intraoperative blood loss in observation group was less and the surgical duration was shorter, but the number of lymph nodes dissection was not significantly different between the two groups. At 3 months after surgery, the lung function parameters FVC, FEV1 and FEV1/FVC in both groups were significantly decreased compared with those before surgery, but the decreases were relatively small in observation group, which had significant differences compared to control group. Serum levels of PGE2, Cor and CRP in the two groups at 1 month after surgery were significantly increased compared with those before surgery, but the increases in observation group were relatively small compared with those in control group. The levels of CEA, CYFRA21-1 and SCC-Ag in both groups was significantly reduced compared to before surgery, and the levels in observation group were lower than those in control group. At 2 years after surgery, metastasis was less in the two groups, and there was no significant difference in disease-free survival between the two groups. In addition, the complications revealed no obvious differences between the two groups. Conclusion Compared with lobectomy, single-port thoracoscopic segmentectomy in the treatment of NSCLC can better relieve the trauma response, maintain the lung function, and reduce the risk of tumor recurrence and metastasis.

The correlation between morphological characteristics of retinal blood vessels and cerebral collateral circulation and prognosis in patients with acute minor stroke
Objective To analyze the correlation between the morphological characteristics of retinal blood vessels and the collateral circulation function of the pia mater, and to explore a predictive model for collateral circulation function and prognosis in patients with acute ischemic stroke. Method 53 patients with acute ischemic stroke were included and evaluated for collateral circulation score using head and neck CTA examination. They were divided into good collateral group and bad collateral group; Using the 90 day modified Rankin score as a prognostic indicator. Analyze fundus color photographs and extract morphological parameters of retinal blood vessels, including the main and branch diameters of retinal arteries and veins in each quadrant, curvature, balance, vascular tree classification dimension, geometric dimensions of the optic cup and disc, etc. Analyze the correlation between parameters with significant inter group differences, collateral circulation score, and stroke prognosis score, and establish a regression model for further validation. Result Among the cases included in this study, the good prognosis rate of the good collateral group was significantly higher than that of the bad collateral group. Among the morphological parameters of retinal blood vessels, the central retinal artery equivalent [(43.29±5.02) vs (39.05±4.21), P=0.002] and average arterial diameter [(51.49±5.35) vs (49.07±4.29) , P=0.040)] showed significant differences between groups. There was no significant difference in the other parameters. The central retinal artery equivalent and average arterial diameter were significantly correlated with the pia mater collateral circulation score, with correlation coefficients of 0.418 (P=0.002) and 0.290 (P=0.035) , respectively. There was no significant correlation between other parameters. The logistic regression analysis of collateral circulation score showed that CRAE was correlated with collateral circulation score (OR=1.349, 95% CI 1.101-1.653, P=0.004) , and the area under the ROC curve predicted by CRAE was 0.747 (95% CI 0.615-0.879, P=0.002) ; The mean arterial diameter was not statistically significant in the regression analysis of collateral circulation score (OR=1.086, 95% CI 0.976~1.209, P=0.129) . All retinal vascular parameters were not statistically significant in the 90 day modified Rankin score logistic regression. Conclusion In mild stroke patients, the central retinal artery equivalent is significantly correlated with the pia mater collateral circulation function, which can reflect the state of cerebral collateral circulation function. The morphological parameters of retinal blood vessels tested in this study cannot predict the prognosis of ischemic stroke.
