蔓荆子总黄酮阻断FoxM1-MMP2 信号轴抑制SMMC-7721源性肝癌干样细胞迁移和侵袭

  • 目的:研究蔓荆子总黄酮(FVTF)对SMMC- 21 源性肝癌干样细胞(LCSLCs)迁移和侵袭能力的影响及其作用机制。方法:体外干细胞培养基悬浮培养得到第2 代SMMC- 21 源性球细胞用作LCSLCs,用溶媒(0.1%DMSO)做对照,不同浓度FVTF 处理LCSLCs,运用细胞迁移试验、Transwell 小室侵袭试验检测FVTF 对LCSLCs 迁移和侵袭能力的影响,采用Western bolt 分析FoxM1 和MMP2 蛋白,探讨FVTF 发挥作用的机制。

Inhibition of Migration and Invasionin SMMC-7721-derived Liver Cancer Stem-like Cells by FructusViticis Total Flavonoids via Blockage of FoxM1-MMP2 Signaling Axis

  • Objective To study the effect and mechanism of fructusviticis total flavonoids (FVTF) on the migration and invasion ability of SMMC-7721-derived liver cancer stem-like cells (LCSLCs). Methods The second generation of sphere forming cells (SFCs) obtained from SMMC-7721 cell line using suspension culture with the stem cell conditioned medium were used as LCSLCs. LCSLCswere treated with 0.1% DMSO or different concentrations of FVTF. The effects of FVTF on migration and invasion of LCSLCs was determined using cell migration assayand transwell invasion assay. Themechanism of FVTF action was#br# explored by analyzing the level of FoxM1 and MMP2 protein expression with Western bolt.

摘要

目的:研究蔓荆子总黄酮(FVTF)对SMMC-7721源性肝癌干样细胞(LCSLCs)迁移和侵袭能力的影响及其作用机制。方法:体外干细胞培养基悬浮培养得到第2代SMMC-7721源性球细胞用作LCSLCs,用溶媒(0.1% DMSO)做对照,不同浓度FVTF处理LCSLCs,运用细胞迁移试验、Transwell小室侵袭试验检测FVTF对LCSLCs迁移和侵袭能力的影响,采用Western bolt分析FoxM1和MMP2蛋白,探讨FVTF发挥作用的机制。结果:与对照组相比,FVTF能下调FoxM1和MMP2蛋白表达,并显著抑制LCSLCs迁移和侵袭能力,且呈浓度依赖性。结论:FVTF有效抑制SMMC-7721源性LCSLCs的迁移和侵袭能力与阻断FoxM1-MMP2信号轴相关。

Abstract

Objective To study the effect and mechanism of fructus viticis total flavonoids(FVTF) on the migration and invasion ability of SMMC-7721-derived liver cancer stem-like cells(LCSLCs). Methods The second generation of sphere forming cells(SFCs) obtained from SMMC-7721 cell line using suspension culture with the stem cell conditioned medium were used as LCSLCs. LCSLCs were treated with 0.1% DMSO or different concentrations of FVTF. The effects of FVTF on migration and invasion of LCSLCs was determined using cell migration assay and transwell invasion assay. The mechanism of FVTF action was explored by analyzing the level of FoxM1 and MMP2 protein expression with Western bolt. Results Compared with the control group,FVTF could downregulate the expression of FoxM1 and MMP2 protein and significantly inhibited the migration and invasion of LCSLCs in a concentration dependent manner. Conclusion FVTF effectively inhibited the migration and invasion ability of SMMC-7721-derived LCSLCs is associated with blockage of FoxM1–MM P2 signal axis.
