Analysis of Fatty Acid Profiles in Children with Severe Obesity Based on On-line Derivatization-Gas Chromatography-Mass Spectrometry

Analysis of Fatty Acid Profiles in Children with Severe Obesity Based on On-line Derivatization-Gas Chromatography-Mass Spectrometry

Abstract

Objective To establish a method for analyzing the composition of plasma fatty acid profiles in overweight and obese children based on online derivatization-gas chromatography (GC) -mass spectrometry (MS). Methods After extraction of 50 μL of children's plasma samples, 50 μL of derivatization reagent TMSH was added. The GC injection temperature was 320℃ and the residence time was 3s to achieve the instant derivatization of the derivatization reagent and fatty acid. Results A total of 11 fatty acid components were detected, and the relative standard deviations (RSD) of their relative contents were all within 20% (n=5). The plasma fatty acids of the children population were mainly composed of C16: 0, C18: 0, C18: 1n-9 and C18: 2n-6, accounting for more than 90% of the total fatty acids. Compared with normal children, the relative contents of C20: 2n-6 and C20: 3n-6 in the plasma of obese and overweight children were higher, and the relative contents of C14: 0 and C20: 3n-6 in the obese group were also higher. Compared with the overweight group, the levels of C16: 0 and SFAs in obese children were higher, while the levels of C18: 2n-6, n-6 PUFAs and PUFAs were lower, and the differences were statistically significant. Conclusion The online derivatition-GC-MS method is simple, rapid and accurate, which provides a new analytical method for the analysis of fatty acid profile in plasma. There are differences in the composition of plasma fatty acid profiles in children with different overweight and obesity status, which may be used as biomarkers for overweight and obesity in children.
