摘要

目的:明确环状RNA 0629(circRNA_0629)在胆囊癌(gaubladder carcinoma,GBC)患者中的临床意义及其与预后的关系。方法:采用生物信息学分析GSE100363与GSE121714芯片结果,筛选出可能与GBC转移相关的circRNAs。收集110例GBC患者的癌组织与癌旁组织,采用实时荧光定量PCR(qRT-PCR)验证circRNAs的表达水平。采用卡方检验分析circRNAs表达与临床病理特征的关系。采用卡普兰-迈耶(K-M)法和时序检验分析circRNAs表达与总体生存期(OS)的关系。采用Cox比例风险回归模型分析GBC预后的影响因素。采用受试者工作特征(ROC)曲线评估circRNAs诊断GBC肝转移和TNM分期的曲线下面积(AUC)值。结果:共筛选出3种circRNAs,分别为circRNA_049637、circRNA_402458、circRNA_0629。癌组织中circRNA_0629表达较癌旁组织升高。癌组织中circRNA_049637和circRNA_402458表达与癌旁组织比较,差异无统计学意义。circRNA_0629表达与肝转移以及TNM 分期相关。高circRNA_0629表达GBC患者1年和2年OS较低circRNA_0629表达GBC患者降低。高circRNA_0629表达、肝转移以及Ⅲ~Ⅳ期是GBC患者预后不良的独立危险因素。circRNA_0629对GBC肝转移和TNM分期具有较高诊断价值(AUC:0.929和0.835)。结论:circRNA_0629在GBC中表达上调,是预后不良的独立危险因素。circRNA_0629表达可作为协助评价GBC肝转移和TNM分期的一种新型标志物。

Abstract

Objective: To determine the clinical significance of circular RNA 0629(circRNA_0629) in patients with gallbladder cancer (GBC) and its relationship with prognosis. Methods The results of GSE100363 and GSE121714 microarrays were analyzed using bioinformatics to screen for circRNAs that might be associated with GBC metastasis. Cancer tissues and paracancerous tissues from 110 GBC patients were collected, and the expression levels of circRNAs were verified by quantitative real-time PCR (qRT-PCR). The relationship between circRNAs expression and clinicopathological characteristics was analyzed by chi-square test. The association between circRNAs expression and overall survival (OS) was analyzed by Kaplan-Meyer (K-M) method and Long-Rank test. The factors that affecting prognosis of GBC were analyzed by Cox proportional risk regression model. The area under the curve (AUC) values of circRNAs for the diagnosis of GBC liver metastases and TNM staging were assessed by receiver operating characteristic (ROC) curves. Results A total of three circRNAs were screened, including circRNA_049637, circRNA_402458 and circRNA_0629. The expression of circRNA_0629 was higher in cancer tissues than that in paracancerous tissues. The differences of circRNA_049637 and circRNA_402458 expression in cancer tissues were not statistically significant when compared with paracancerous tissues. The expression of circRNA_0629 was correlated with liver metastasis and TNM staging. The 1- and 2-year OS were lower in GBC patients with high circRNA_0629 expression than those of GBC patients with low circRNA_0629 expression. High circRNA_0629 expression, liver metastasis and stage III~IV were independent risk factors for poor prognosis in GBC patients. circRNA_0629 had high diagnostic values for GBC liver metastases and TNM staging (AUC: 0.929 and 0.835). Conclusions circRNA_0629 expression is upregulated in GBC and is an independent risk factor for poor prognosis. circRNA_0629 expression may serve as a novel biomarker to assist in the evaluation of liver metastasis and TNM staging in GBC.
