DFMG对内皮细胞COX-2 表达和黏附功能的影响

摘要

目的:评估7-二氟甲氧基-5,4'-二甲氧基金雀异黄素(7-difluoromethoxy-5,4’-dimethoxygenistein,DFMG) 对损伤的人脐静脉内皮细胞( HUVE-12)中环氧化酶2 (cyclooxygenase-2, C0X-2)蛋白表达的影响及对HUVE-12细胞氧化损伤的保护作用。方法:肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)诱导建立 HUVE-12损伤模型,不同浓度的DFMG对其进行诱导;流式细胞术(Flow Cytometry,FCM)检测细胞凋亡、Western blotting检测COX-2蛋白表达、蛋白定量分析法测定内皮细胞/单核细胞的黏附率、ELISA检测E-选择素、单核趋化蛋白-1 ( Monocyte chemoattractant protein-1, MCP-1)的释放。结果: DFMG 能拮抗损伤的内皮细胞活性的下降,并呈浓度依赖性,同时DFMG也能呈浓度依赖性地下调COX-2蛋白的表达、下调内皮细胞与单核细胞的黏附率。结论: DFMG可能通过减少COX-2蛋白表达水平而减少单核细胞/内皮细胞黏附作用,进一步减少炎症渗出,拮抗炎症反应,保护血管内皮细胞。
