Abstract Objective To investigate the effect of nerve growth factor (NGF), transforming growth factor β1(TGF- β1), fibroblast growth factor (FGF) in fracture healing. Methods Selected 30 male New Zealand rabbits, were randomly divided into 5 groups, including saline group, NGF group, TGF-β1 group, FGF group, NGF+FGF+TGF-β1 group, 6 rabbits in each group, tibial fracture model was established in all animals. The saline group, NGF group, TGF-β1 group, FGF group, NGF+FGF+TGF-β1 group after operation in the fracture site were injected saline, NGF, TGF-β1, FGF and NGF+FGF+TGF- β1, observed each group the X-ray score, bone mineral density and osteocalcin levels. Results Postoperative 14d, 21d and 28d, NGF group, TGF-β1 group, FGF group, NGF+FGF+ TGF-β1 group X-ray score, bone density and serum osteocalcin were significantly higher than those in the saline group; Postoperative 21d and 28d, NGF+FGF+TGF-β1 group X-ray scores were (15.63±0.92) and (16.01±0.93), significantly higher than that of NG group F, TGF-β1 group and FGF group; Postoperative 14d, 21d and 28d NGF+FGF+TGF- β1 groups bone density were (0.604±0.019) g/mm2, (0.613±0.021) g/mm2 and (0.644±0.016) g/mm2, significantly higher than that of NGF group, TGF- β1 group and FGF group; Postoperative 14d, 21d and 28d NGF+ FGF+ TGF-β1 groups serum osteocalcin were (10.236±0.121) ng/ml, (11.924±0.093) ng/ml and (13.210 ±0.083) ng/ml, significantly higher than that of NGF group, TGF-β1 group and FGF group. Conclusion NGF, TGF-β1 and FGF has a certain role in promoting fracture healing, them has a strong synergistic effect, can obviously promote fracture healing.
